I recently installed Win10 64-bit onto my desktop after getting a new SSD (the OS is on the SSD, so everything is a fresh start [my old HD is unconnected] ). For some reason, the USB's on my MoBo randomly stop working. I know this because my keyboard, mouse, and wifi are all USB and they randomly go out and don't come back until after a restart.

Solution
1| Have you made sure your BIOS is up to date?
2| Have you installed the latest drivers for your motherboard, namely the chipset and USB drivers?
ASMedia® USB 3.0 controller :
2 x USB 3.0/2.0 port(s) (2 at back panel, blue)
AMD SB950 controller :
12 x USB 2.0/1.1 port(s) (6 at back panel, black, 6 at mid-board)
ASMedia would need latter drivers(although which chip was used is not stated both in spec's or in the manual) while the chipset drivers from AMD are found here(provided you're running on a 64bit OS).

3| Try and run the USB 3.0 drivers found for older OS in Asus's support page and run them in compatibility mode:
Right click installer>Properties>Compatibility Tab>Windows 7/8...

You should also disable automatic device driver updates feature and install necessary device drivers manually.

Lastly, try a repair install and see if the issue is alleviated.

Can you please pass on your full system's specs inclusive of your connected peripherals?
