Question ASUS Prime Z490-A Unlabled headers

Does anyone happen to know what these 2 headers are? They are not mentioned in the manual at all. I'm sure they are for ASUS service and repairs, but would like to know what bus or protocol they use (LPC maybe?) and how do they use them to provide service.

Can you please parse a picture of what you're looking at? Host your images on a site like Imgur and pass the link to this thread for us to see and hopefully identify it. To add, if the header isn't labelled on the manual, it probably due to the fact that it's not of any need by the end user. The fact that you don't know it, means that it's not needed for your everyday use case.

Then again, I've seen people mislabel/misinterpret ports during a build.
 
What headers are you talking about? Usually when you say "these" it is followed by some image or explanation that specifies what "these" are.

The first on is just behind the M2 (WIFI) slot. It's a 2 x 5 with 1 pin missing.

The second is next to the power button on the board. It's a 2 x 5 with 1 pin missing.
 
Neither of them are listed on the board diagram nor in the user manual, so they are almost certainly either trusted platform device headers or diagnostic and service headers used by the manufacturer for initial configuration or diagnostic service. With no other information available, it's really just a guess. There are any number of things those two headers might be intended for but it's likely they are only useful in a factory environment using the specific required equipment to make use of those headers.

Beyond that, no idea.
