Solution

I also have an ASUS motherboard Z170 Mark 1 running under Win7.
I have a Samsung NVMe SSD 950 pro 512GB inserted in the M.2 motherboard slot.
Win7 is installed on this SSD NVMe and it boots fine (bios version 1902).
I also have...

maybe the slot is shared with the m2 ?

try in another slot and see .

will research it soon

I also have installed the ASUS Hyper M.2 PCI-E Mini Adapter Card, that was delivered with the ASUS Mark1 board, into expansion slot PCIEX16_2 (second PCIE slot from the bottom of the board). Into this card I have inserted also a Samsung NVMe SSD 950 pro 512GB for storing data.
Works great. Fast. Checked performance with Samsung Magician (newest version required!).

I have a second PC running with the same equipment.
Only difference is the lower/older Bios version of 1801.
Works also fine - booting from M.2 slot.

Of course you have to select the correct SSD in the boot order of the bios setup!

Solution
Last pci express will disable on board m2 slot when using adaptor on that slot. Also its slower. Use pci express next to tge gpu which will give u full speeds and compatibility

... that is exactly the second PCIe slot from the bottom ...
