Yes of course but it can be easy or not so easy.

If you are running Windows 7 you can shrink your C volume then make a new partition in the empty space. The Computer Management/Disk Management tool does this. I'm sure you can also do this using the command prompt. diskpart I think.

You can always make a drive image using Norton Ghost, Acronis True Image or some other such program, clear the drive off, make new partitions, and reinstall the image. I do all this using the Norton Ghost recovery disk. Not trying to advertise just saying how it can be done. Only way to go if you are playing with a RAID array and booting from it. I have reconfigured my RAID array more times than I can remember and was booting from it.
