Can I connect my monitor to my motherboard?

I have 3 1440p monitors but my gpu only had 2 displayports. My motherboard has a displayport on it. Will it work ?

That depends on your bios of your motherboard. Do you have the option in the bios to connect it to both a add-on card and the motherboard? Most motherboards make you pick one.

About all modern boards disable the boards graphics when a video card is used.
Since you already have it try it and see.
Most monitors and video card have more connections than just displayport.

Unless the monitor has HDMI 2.0, no.
