my config is AMD FX4100 on asus M5A88-M with 8 GB RAM(it has 4250 built in GPU.
Now i want to upgrade with SAPPHIRE DUAL-X 14-202-099 Radeon R9 280 3GB 384-Bit GDDR5 PCI Express 3.0 CrossFireX Support Video Card
You will be bottlenecked by that CPU. You should get an FX-6300 and if you don't have dual channel, get it. For the GPU, that is fine. Yes, it will fit.

You will be able to run that GPU in a PCI-E 2.0 x16 slot. A note to have is that PCI-E 3,2 and legacy all are intercompatible if you put a newer version in an older.
Don't expect the full performance of it though due to CPU bottle necking. Otherwise you will do well.
