I'm doing a build using the Asus Z-170 Deluxe Mobo, and I plan on running two nvidia GTX 1080 founders editions in watercooled sli as well as 32Gb of RAM. I have heard great things about the speeds of the M.2 Especially the samsung 950 pro. A lot of people will immediately answer this with, "You dont need it, just use a smaller Sata 3 ssd for your OS and a larger one for your data. But I WANT to use an M.2 for my OS. I will also be using two samsung 850 pro 1tb ssd's, but i would like to utilize an m.2 for my os rather than another Sata III, purely because i want to utilize m.2 and only because I want to. So all I need to know is if it will work with 2 1080's in sli and if so will it cut into their performance?

M.2 port gets its lanes from the chipset usually. Z170 cpu''s only have 16 pcie3 lanes for the gpu's so if you are happy running your two 1080's in SLI at x8 each then you should be fine otherwise you will need to upgrade to x99 and a 40 lane cpu. With as powerful as 1080's are, I have not seen any benchmarks noting how much performance is lost in x8 sli. Previously it was minimal, a few frames.

But arent the PCIE lanes on a X99 2.0 whereas Z170 is 3.0? Because if so, there should be relatively no difference between x16 x16 sli on 2.0 and x8 x8 on 3.0, correct?
