I have a Samsung EVO 500 gb SSD and a Sandisk Ultra II 480 gb SSD. I have Win 10 on both. Can I switch them at bay 0 semi-regulary and boot both with no problem?

Solution
If this is not a hot swap bay and you shut down to swap out the drives then it may work. You may need to enter the bios and make sure the new drive is in the boot order if you've never booted from it before.
