[SOLVED] Can I transfer data between two internal hard drives that are in use?

I have a 500gb HDD that I want to transfer to a 2tb HDD and both of them have data on them. Can I transfer data without formatting?

As long as there are no free space constraints (such as needing to copy 400 GB of data to a drive with only 200 GB of space remaining, etc), copy/paste/transfer away...

And as long as the "data' isnt really a program...
Programs usually install stuff to the Windows registry and the users hidden Appdata folder, which includes their location.
