Depends what it was. If it was an app, it'd stop working because it would think it was on the old drive. Data - nothing - you'd just have to remember where it was when you needed it.

The program would stop working. Registry settings tell the program where to find it's files/executables/drivers. If you want to change folders, you'll need to re-install the program.
