Does it matter if i use the blue or black pcie lanes.

Will i lose any performance if I use the blue.
Atm its plugged into the blue port.
Motherboard is : P8P67 PRO (REV 3.1)

according to motherboard specifications, black pcie port is limited to 4 lanes maximum.
so using blue is recommended.

As general rule with most, the pcie lane on top (closest to processor) is best choice for graphics cards.
