I am looking for a cable that will connect eSata to mini USB port. Looking at this cable on Amazon, it has an extra eSata head on the end of mini USB, would it work still work if the extra eSata head is unused, or is there some sort of special port that requires plugging combination of the eSata and mini USB heads simultaneously on one end?

I don't know what device you have.
But you need to know the eSATA cable only can transfer the data, but not provide the power. The mini USB port can provide the 5V power and tansfer the data too.
If you have a device like camera with the mini USB port, then you can just plug the mini UBS into the camera, the other end connect to the PC eSATA port. So you can use it without use the extra eSATA connector.
If you have the 2.5" external HDD then you can use that cable too , but not for the 3.5" HDD because it requires 5V and 12V to power up.
