should I adjust them in the NVIDIA control panel , or in game settings ? what if I have tipple buffering in the NVIDIA control panel as on and in the game as off , will it be on or off in the game.

It will be off - if you disable it in the NVIDIA control panel it will not be enabled if you enable it in the game. It's better to adjust them in game but if you need to, you can enable them in the NVIDIA control panel and then in game.

The things you do in nvidia control panel will usually override the in game settings (depending on the game) generally i would say leave everything to application controlled and adjust your settings in game. Nvidia control panel is good if you want to force a certain kind of anti aliasing or use adaptive vsync.
