GeForce GTX 750 1GBDDR5 compatibility with ASUS B85M-E mATX

I understand backwards compatible, but I am trying to "min-max" my new build, so should I be upgrading the mobo to one with two PCIe 3.0 slots or find a different GP?

Currently Graphics cards can't even use all the bandwidth of even PCIE gen 2.0, so going from a pcie 2.0 to pcie 3.0 will yield the exact same frame rates.

You will see no difference between pcie 2.0 vs 3.0 with your GPU and motherboard. They are backwards compatible, so dont worry.
