Graphics Card/Motherboard Advice?

Wanted to extend my PC's life a bit longer to play games on. Replacing a Radeon 5770. Will an Asus P7P55D-E board accept a Radeon R9 270X?

That Graphics card will work on that motherboard just fine, as it has a PCI-E X16 slot. Don't worry about the GEN 2.0 slot, as PCI-E is backwards/forwards compatible.

Yes your mobo should be OK. Hopefully you've got a good CPU, room in your case and a power supply that can run the 270x.
