Hard Drive performance

What do I look for (what kind of tests i.e Write Access, Read Acess?) in the performance charts from Tom's Hardware that will cause a huge improvement on performance between two hard drives? I am trying to decide which hard drive to install my OS on.

I recently bought the WD Scorpio Black 7200RPM 750GB 16MB Cache 3Gbps.
Old hard disk would be WD1600BEVT 160GB 3Gbps 5400RPM but it was not on the chart so I put the two closest models, WD1600BEVS and WD3200BEVT

Goal: Determine which to install OS on. Will it benefit from a HUGE performance increase? ~>50%?

Agree with the above poster - and if you want to see some graphing and quantitative results for comparison, download and run "HDTune"

HDTune will also give you some additional information regarding the hard drive's health.

The two drives you listed have different RPMs - 5400, 7200 - generally the faster RPM drive will perform better.
