Yes.
There are 3 basic thicknesses of 2.5" drives.
7mm, 9.5mm, and a few 15mm (large capacity HDD).

The spacer is to ensure a 7mm will fit in a laptop built for a 9.5mm, and have it not move around.
Some drives come with the spacer, to use if needed.
