Solution
OK my friend, I got the full picture now. Absolutely get the USB 3.0 card for your needs. You have external hard drives and a very fast thumb drive -- all of them need USB 3.0 and you have a super slow USB 2.0 mobo. The speed will be at least 3x faster for your thumb drive and at least 2x - 3.5x faster for external hard drives (depending on which model you have). Here's a card with a good rating that fits your mobo:

You can also add this if you want the USB 3.0 connectors on the front panel of your computer (make sure you have a 3.5" bay)

If you only...
I'm not finding anything about your mobo online, but if it is PCIE 2.0 it will be a bit slower than USB 3.0. If your mobo is PCIE 3.0 then it will be faster than USB 3.0 and the card will be able to go as fast as it can. In reality, you never get max throughput of the USB interface due to overhead and the quality of the interface and/or the type of media of the connected device. For example, most USB 3.0 thumb drives will not be anywhere near the max possible advertised speed of the USB 3.0 interface. You should be fine even if you have PCIE 2.0 or PCIE 3.0.

No AMD boards other than AM4/TR4 support PCI-Express 3.0, so its most likely a 2.0 slot.

Ok, so it’s about whether you want to go from USB 2.0 to a 80% USB 3.0, which means 60 MBps to 500MBps. Just get the card. It’s a huge increase in speed, even if the PCIE slot doesn’t max out the USB 3.0 card. USB 3.0 is 640MBps, so like I said, 500MBps is still plenty fast

i read about PCI Express sata 3 USB 3.0
a board have 2 port USB 3.0 and 2 SATA 3 port

my motherboard support sata 2 and i want to know what is the i wrote above ?
then which one is useful for me ?
i have 2 HDD External USB 3.0 and One usb 3.0 Flash Drive
i want to transfer file as fast as possible.

I thought you wrote that you found a board (pcie card) that has both USB 3 and SATA 3. Sorry if I misunderstood. I think you are asking if you should get a USB 3 card or a SATA 3 card, right?

I have an old computer without USB 3 and I bought a USB 3 card for it. I also bought an adapter that has USB 3 to SATA 3 for 2.5” drives (5 watts max). StarTech model USB3S2SAT3CB and it is around $11 USD. If you need to attach larger size 5.25” SATA drives you have a few options. You could open up your computer case and plug the drive in temporarily to work on it, or you could get a SATA PCIE card, or you could get a SATA dock with USB 3 plug and use it with a USB 3 PCIE card. I think USB 3 card is still the best option because USB 2 is so painfully slow compared to USB 3 and you can still use SATA devices in the various ways I described.

Important things to remember:

You’re USB thumb drives will still be very slow (at USB 2 or less) if they are not USB 3 thumb drives. You must use a v3 thumb drive in the v3 USB port in order to get the USB 3 speed.

Even if you have a SATA 3 drive and you plug it into your existing motherboard SATA 2 port, you will not suffer much loss in speed. The Samsung 850 evo with SATA 3 is 540 MBps, and if connected to SATA 2 it will be probably around 270 MBps or half the speed it should be. But, a USB 3 upgrade will increase your USB speed about 10 times! (and remember you can get a sata adapter or dock that plugs into the USB 3.

Is this the motherboard in question?

I don't see any problem using pcie USB3 card in it to get USB 3 speeds.

If you only have 5.25" bay available, get this for the front panel usb:

EDIT: Bear in mind that your new bottleneck will be your internal hard drive, since it is from 2009 and averages about 125MBps read/write. It is still beneficial to upgrade to USB 3.0 because 2.0 is only max 60MBps (real world performance is probably half that). Your USB 3.0 devices will absolutely benefit from the upgrade.
