I currently am having my computer built and I was wondering if the HDD I chose would really have an effect in my situation and how much if so. As I am using it for Professional Video and audio Editing
I currently have a 950 Pro Series 512Gb m.2 SSD for my OS and any important applications, then will have 2 Seagate 4TB 5900RPM HDD's For storage and one for Backup, Will this really have much of an effect for when storing and backing up larger video files? and will this at all effect my rendering and use of video files? I have dedicated to atleast one of the 4TB HDD's but the second i can get a 7200RPM HDD if it would make a difference, and would I want that as main storage or backup?

It should be fine as a backup and storage drive...

It should be fine as a backup and storage drive, if you aren't booting from it or using it as an OS. My 5400rpm drive works perfectly, so your 5900rpm drive should work great.
