This motherboard that I'm planning to get for a pc build in the future only has one fan header at the top right corner for front fans. I'm also not sure if the case will have good airflow.

Here's the link to the case: https://www.amazon.com/Cooler-Master-MasterBox-Mid-Tower-Brushed/dp/B076BXJ76J/ref=sr_1_3?dchild=1&keywords=mid+tower+pc+case&qid=1608401654&sr=8-3&th=1

Would one front fan and rear fan be good enough for optimal airflow through the case? Or do I need a fan splitter in order to have two front fans?

It CAN work - in specific setups, such as cpu/apu only, or systems with low power cpu and gpu.

The MB600L looks like a 'hot box':
-'Sealed' front intake.
-No options to install fans at the top for enhanced cooling.
So, it's a front to back airflow chassis, but the front intake is choked.

Well crap, look's like i'll be reconsidering my case then. The build will be a cpu only for while, but then later on I can get a graphics card that will require good airflow.

I'll look for another one now, and send the link here.

Edit: How about this one? https://www.amazon.com/Antec-VSK10-...ATVPDKIKX0DER&rnid=303116011&s=pc&sr=1-8&th=1

-'Sealed' front panel.
-Dual slot gpus and larger will be hurt by the proximity to the psu shroud. Blower gpus can get around that restriction though.
-Acrylic side window can bend/warp when exposed to enough heat.
+Well, you've got top panel cooling options now.

Slightly better than the last one - just slightly.
