Question Is the PCI slot broken?

Hello mates! I recently bought an used PC. The gpu was installed in the second pci slot and i wanted to move it in the first slot because I was aware about the fact that it can limit the potential of my gpu. When I moved to the first slot and i connected everything to my monitor, it didn't give any signal to it. Therefore I changed to the second PCI slot and It works all fine...I am really worried about the fact that the main slot is f ed up...
Motherboard: Asus B150 pluse
GPU RX580 8gb
I know that the graphics card supports PCI-Express x16 3.0 but it is currently running at PCI-Express x4 1.1
