I have the following psu: colorsit en60950 500w, then it says +3.3V 28A, +5V 35A, +12V 25A, -5V 0.5A, -12V 0.8A, +5VSB 2.5A
Will it be enough to run the R9 380?

That depends, with 15.8a required for the card you have less then ten for everything else.
This is the rest of my rig:
i3 2100
h67 motherboard
4gb ram
1 500 hdd
1 dvd writer/reader
You know actually i have two psu, one is the stock HP psu that came with the pc itself, and then i have the colorsit 500w psu. Would it be safer to power up the GPU externally with the colorsit and leave the other components to work with the stock psu?
If you go by the published numbers then your PSU is not up to the task, however as I have had a 380 running flat out on a 400w (currently on a 430w PSU) I would be inclined to give it a go myself.

In theory 25A on the +12V rail should be enough for a 380, although that looks like a pretty old PSU... I'm not sure I'd trust it with a high end card like that....

The 380 is not a high end card as it's comparable with a GTX960 and even though it is more power hungry it can still be run on a 400w PSU.
