So how much of a speed differnce from pci-e 2.0 and Pci-e 3.0,Because I have a Asus m5a97 R2.0, and my PCI-e is 2.0 so will there be a big performance drop? and what about Sapphire Radeon VAPOR-X R9 290 4GB GDDR5 what Pci-e speed does it use?

You won't see a difference, we're talking about 1-2 fps drop MAXIMUM. The Sapphire Radeon VAPOR-X R9 290 4GB GDDR5 uses PCIe 3.0 but you can surely use your motherboard with this GPU without any issues.
