According to MSI yes the MOBO supports both SLI & CF - http://us.msi.com/product/mb/Z87-G45-GAMING.html and by the looks of it with triple spacing you'll have one PCIe width for additional GPU ventilation.

It should work. I'm running Z87-GD65 with GTX760 TF 4Gb on Dual SLI. SLI will only work on the first and second PCIE slots. If you put it in the 1st and 3rd, your SLI cable won't be able to reach it. The 3rd PCIE slot is for triple CFX because nVidia has specified its card to work on minimum 8 lanes. so putting a 3rd nvidia card is basically useless as it's only running PhysX. Whereas AMD works on minimum 4 lanes so you can got triple CFX on the G45
