I am currently using a Pentium G4600 on a Gigabyte B250M DS3H motherboard and I am planning on upgrading to an i5 9400f on a Gigabyte B365M DS3H motherboard. Will I have to do a backup of my system and/or reinstall Windows even though the motherboards are very similar?

In that situation it's recommended to perform a clean installation.
You will be moving a Windows that was configured for a different motherboard with different drivers and settings.

If you still want to try moving Windows without re-installing, before removing the disk, uninstall all drivers (sound, network, chipset, etc.) related to the old motherboard. That way you will have a lower chance of conflicting drivers.
