ntfs or large fat32

I am trying to create a bootable usb drive with rufus. Which of these is better or which should i use?
Also what is the cluster size and which should i choose?(16, 32, 64 kilobytes)

For Windows files over 4GB (because FAT32 and Large FAT32 cannot have files that are larger than this) use NTFS and 4096 bytes (cluster)
