Probably not.

"Probably" only because I do not know how your system is configured. E.g., there may be some apps hosted on D: at startup.

Or other apps looking for data that is stored on D:

Just disconnect D: drive by removing the data and power cables.

Go through a few boot ups, work and game as usual. If all goes well then physically remove D:
