I'm not at all knowledgeble on PC hardware and software, so please try to structure your answer as simply as possible.
I had a new motherboard installed into my PC by a friend and didn't boot it up for approx. 6 months, and now suddenly the audio doesn't work in my headphones, though it does work on the monitor's built-in speakers. I've tried updating the drivers, uninstalling and reinstalling the drivers and checking the default outputs, but nothing seems to work. Could someone please provide a list of solutions before I take my PC to a hardware store?

Right click on the speaker icon down by where it shows you the date and time (It's called the system tray)
Select Playback Devices.
You likely have several selections, try/test them and set the one that works with your headphones as the default playback device.

I'm thinking the default device reverted to the audio that is produced by your GPU.
