I'm using Windows 7 64 bit and have all of the correct drivers installe(removed and reinstalled several times) but my PC can't even SEE the connected 5.1 speaker system.

After a reinstall of windows I can only get sound from the HDMI cable going from my GPU to the screen, says speakers are not connected.

I'm using an onboard sound card from an Asus P8P67-M with realtek HD drivers.

Sorry, another silly question: are they plugged in and turned on?
Good.
Re-boot, open the BIOS and make certain that the on-board sound is chosen. Sorry, I don't recall what the UEFI bios calls it but it should list something about RealTek. Remember that Realtek also runs your LAN/Ethernet, and we don't want to mess with that.
JUst a thought here:
Unplug the HDMI cable from the monitor, unless it is how you are getting your picture/video.
(If this is how you hooked your video card to your monitor it may be the only way to get sound until you unplug it and run a DVI or Display port cable to the monitor)
If you haven't already done it, open the Control Panel, click on and open 'Sound', if your speakers show up (they will be listed as "Speaker" Realtek High Definition Audio), make certain they are check marked as the Default Device. UN-check any HDMI output that is checked.
Rt. click your speakers (still in 'Sound'), click properties, open the Advanced tab, and make sure the boxes under "Exclusive Mode" are unchecked. Click o.k., that will return you to the main sound window, click the 'sounds' tab at the top, choose a sound,
such as Asterik, and test it at the bottom of the window. Hopefully you will have sound.

It's late here in the West, and I have had it for the day. Sorry if you have done all of these things, but you have given us limited info. If you have tried all these, or you do try them, post back please and I will try to help tomorrow. Good night, 'chat' with you tomorrow.

There's no Sound option in the BIOS UEFI menu but I enabled everything Realtek related just in case.
Speakers are "greyed out" so there's no advanced menu and I can't pick them as default.
I can uninstall the HDMI sound drivers if that helps.

I've just using the drivers that came with the MB CD(which worked the first Windows install) and I tried new ones from the asus site, completely removing the drivers each time using driver sweeper but no luck.

Your motherboard manual has the information on the audio and its location in the BIOS; Turn to page 2-21 to read it, but here it is.
Open the BIOS and if it is set to EZ, switch the setting to ADVANCED; This will give you the full menu to work from. Once the BIOS is open in the advanced mode, click on the Advanced tab at the top; in the menu that show up click on 'Onboard Devices Configuration'.
The first choice should be "HDAudio Controller". Make certain it is Enabled. Next choice will be 'Front Panel Type', make certain HD is selected.
Enable VIA1394 ONLY if you have something that uses a Firewire to attach to the PC. Set the PCI Express15_2slot to "Auto".
The JMB Storage controller enables your SATA2 (3Gb/s) plugs on the mobo, if you installed the drivers from the install CD, then enable it. If you didn't install the drivers or are NOT using those ports, disregard.

Until we get the speakers to show up, don't worry about the audio drivers. If you have drivers installed - whether from the original CD or downloaded and updated - they will work once we get the PC to recognize the speakers.
