my question is will it be worth using other drives as cache to make it run faster.
will the cache help render times or faster preview in after effects.

You have 3 x SSD's? What other drives are in this system?
What space is the AE cache on now?

If the drives are NVMe, you should see some performance boosts using them partially as cache, or by using a technology like Intel Optane.
