Will Asus AMD Radeon R7 250X (2GB, PCIe 3.0 x16) fit into and work well with my pretty outdated motherboard Asus P5Q3?

I have the R7 250X 1GB, it fits and works on my ASUS M5A78L-M LX3, which is a microatx mobo with a single PCIe 2.0x 16.
I think it should fit fine on your mobo.
