Hello,
Normally no reboot is required after you have assigned a drive letter.
Possibly the partition is damaged (for example the boot sector), and
possibly - if it is damaged - it can be repaired. The partinfo tool on our
support page can reveal partition table and boot sector damage, you're
welcome to post a report in our support forum. Alternatively a program like
our iRecover can be used to copy data from damaged volumes to another drive.
