SATA Express vs SATA 6 gb/s

Whats the difference between the two? I have 4 sata 6 gb/s slots and 2 sata express slots. Im connecting a 250 gb ssd and 2 7200rpm 1 tb 6 gb/s hard drives. What slot should I connext them too?

Note I also have a cord for a front panel e-sata slot. So techincally I have 4 cords i need to place
