So, I am using Win10 64 bit and having an AMD 6300 graphic card. My driver is up to date. Now my second monitor is detecting when I click on the detect button; it has written: "Didn't detect another display."

Kindly help me please..... the Second monitor is connected through DVI TO VGA Adapter thr graphic card.

To run two monitors you will need to have them BOTH connected to the graphics card. It will not work to have one connected to the graphics card (and so driven by the graphics card) and one connected to the motherboard (and so driven by the processor's on-board graphics)

And you will need to make sure your graphics card is capable of running two monitors at the resolutions you intend.

Of course it is connected in the same graphic card and NOT in onboard. Yes, graphic card is capable to run on 2 monitors.
