Hello, I am thinking of buying another M.2 Disk and I want to know if this configuration works well for me, in total there would be three M.2 Disks and one sata.

Motherboard: MSI MEG Z390 ACE

SSD: Corsair Force Series Gen.4 PCIe MP600 500GB NVMe M.2 SSD
Silicon Power 1TB NVMe M.2
Silicon Power 500gb NVMe M.2
Silicon Power 1TB SSD
In the Bios you can see that I have 5 sata ports and an unused M.2, if I install a third M.2, would the sata ports be disabled?

No not generally it only disables certain ports most of the time and that generally when gen 4 is being used or on some older boards the sata m.2 port
