Two power connectors on Gigabyte GA-M57SLI-S4

Gigabyte GA-M57SLI-S4 REV 2 has 2 power connectors, a 4 pin and a molex connector... do I have to plug in both?

4 pin - The ATX_12V power connector mainly supplies power to the CPU. If the ATX_12V power connector is not connected, the system will not start.

molex - PCIE_12V (Power Connector)
This power connector provides extra power to the onboard PCI Express x16 slot. When installing two graphics cards, please connect the power cable from the power supply to this connector, or system instability may occur.
