Most of the SSDs and HDDs have support for SATA3 Only...

And most of the external HDDs support USB3.0.....

What is the purpose of the SATA Express ports on my Motherboard and what can I do with them ?

SATA express was a failed interface. No significant storage devices were ever made. Just ignore the ports.
