I recently got GIGABYTE GA-Z170X-Gaming 5, formatted the PC and installed Win10.
I got the Soundblaster from the CD that came with the motherboard

I am quite unfamiliar with audio drivers and sound devices , so I apologize for my limited knowledge.

I get this message whenever I open the XFI MB3 controller:
"The current selected audio is disabled, not present, not supported by the application, or has unplugged jack connections. Do you want to select another audio device now? Yes or No"

When I select No, it opens up but the control are greyed out and doesn't detect my audio devices. If I choose yes, a window shows and let me choose which device - Windows Default, Realtek Output Optical, and Realtek Output.

This message also appears whenever I plug an audio device to the back panel while the application is running.

I suspect it might be conflicting with Realtek, but I've read the the Motherboard has a built-in "Realtek ALC1150 115dB SNR HD Audio" and I assume it would work with SoundBlaster XFI MB3. http://

Solution
There is a resources & driver conflict between the two sound devices.
Disable the integrated (on board) audio device in the system BIOS settings (BIOS Setup).
That will render the Realtek sound adapter inoperative so there will no longer be a conflict.

If you can't find where to disable it in the BIOS settings, disable Realtek Audio device in the Windows Device Manager instead (right-click & choose 'Disable'). Then restart Windows.

I had the same issue, reinstalling the Realtek Drivers didn't change anything, i had to uninstall the creative X-Fi software, restart my pc and reinstall that and everything worked fine, some files got corrupted somehow.
