Win2008 two networks card

I have two NIC card. NIC1 and NIC2. NIC2 is configure for my LAN and all works fine. I need to create separate network for my backup software to use. As soon i bring NIC1 online i can't access the server on NIC1.
here my configuration:
NIC2:
IP: 10.1.15.55
Mask:255.255.255.0
Gateway:10.1.15.1

NIC1:(backup network)
IP: 10.1.21.10
Mask:255.255.255.0
no gateway

As soon I enable NIC1 i cant ping the server on address 10.1.15.55 but I can ping from the server any machine on the network.
Any help fixing this issue
 
I assume you can ping stuff in the .15 network and the .21 network from this server.

Go to a cmd prompt and do a route print and verify that you only have a single default route to 10.1.15.1. You can also check your ARP tables in various places to ensure you get the correct mac addresses corresponding to the ips.

Your design is correct and is "should" work.

the routing table looks fine. there is only one gateway. the odd part is that other computer on the network can't see this server. when i ping it there is no replay. I do have 2003 one with the same configuration and is working fine. any ideal please
 
Look in the arp table for both machines to see if they have correct mac addresses. You would have to run a packet capture to see if they are even receiving the ARP requests.
This type of thing normally would be a vlan issue if you are using a switch between or worse a bug in the server firmware. Sometime the offload feature causes strange errors
