SOLVED!
Solution: At the same time as you change the Displayport monitor to Extend desktop to this display make sure to check the Make this my main display. If you run Displayport in a multiscreen setup Displayport has to be the main display.
My displayport monitor suddenly decided not to work, after having worked perfectly for more than a year. I just started up my computer one day and it didn't work. The same monitor plugged in via HDMI works without any troubles.
The strange thing is that the displayport monitor is discovered by windows, both in NVIDIA Control Panel and in Display Settings, but if I try to enable it it causes problems.
What it looks like in Display Settings (1 is the displayport monitor, 2 is the same monitor using HDMI, both are currently plugged in. The problem is still there with a different monitor using HDMI):
If you change the "Disconnect this display" to "extend to this display", you get this error:
Here is the same thing in NVIDIA Control Panel:
Philips PHL 242G5(1 of 2) is the displayport version, (2 of 2) is the HDMI. If you enable the Displayport version (with and without the HDMI plugged in) the computer freezes for a few seconds, then removes the check from the box without displaying any error.
I have tried reinstalling Windows to no avail.
