Windows 10 installation from used USB

Several months back I built my PC and bought the windows 10 USB from Best Buy. Fast forward to now, my little brother built his PC and needs Windows 10 as well. Unfortunately, because all the stores are out of the Windows 10 software, and we need this computer to be functional very soon, my question is, can I use my USB that I used already to install Windows 10 on the new computer, and then buy the product key somehow from Microsoft? If so, can you please link me the URL? If not, is there any other viable option out there to get Window 10 on the new computer?

Because of the new Anniversary Edition of Windows 10, it would be MUCH MUCH FASTER to download a copy of the new ISO and install using that. (You still have to buy a key from Microsoft.)
