"Windows Easy Transfer" File is Located at the disk that i'll install 64bit , would it be still there when i format it and go to 64bit from 32bit? or would it be also deleted?

it is a 1tb disk and windows easy transfer file is 240gb.

Formatting a disk will erase the contents, yes. Copy the WET file to a secondary disk first.

Generally speaking, if you format the disk then any content on the disk would be lost.

You can copy that folder off to a removable storage device and then copy it back once your upgrade is complete.
