How do I make a non bootable Vista ISO file bootable for virtualbox? I downloaded the file from microsoft.com and don't want to accidentally boot it on my actual PC. Thanks

Given a valid ISO file, in VirtualBox, you just have to tell it where that ISO is during the "install" of this OS.
Your PC won't try to boot from it.

I thought so too but I just got a "No bootable medium found" error message on startup.

Startup of what? The VM you wish to designate Vista to?
 
In your VM settings you should be able to add a CD/dvd drive -mount Iso there boot the VM and install windows.
second possibility is to apply the ISO image directly to the virtual hard disk file the VM uses -you can use gimagex for that, for example:
