I have a Z690 A pro motherboard with 2x8GB memory. I want to upgrade my RAM to 3 x8GB RAM. Is it possible to use 3 slots?

Yes but it will slow down the whole RAM department because the 3rd stick is not running in dual channel mode.

I cant use the 4x8GB because my AIO blocked the fourth ram slot ...

If 32GB RAM is a must, you can invest in 2 ticks of 16GB each.

There is also no guarantee that mixing a new stick with your 2x8gb will work without issues. RAM is best bought and used as matched kits.
