I have a laptop, with 4GB soldered Ram probably single ranked.
Will it work if I add a 16GB dual ranked to the extra slot? as I can't find a 16GB single ranked

Solution
What is the make/model of your laptop?
Go to a ram site like Kingston or Crucial and enter your laptop in their ram upgrade app.
You will get a list of supported upgrades.

Likely, you can install a 16gb module resulting in 20gb of ram.
8gb will be running in dual channel mode and the remaining 12gb will be in single channel mode like you are now running.
Overall, a very good upgrade IF possible.

Solution
Run cpu-Z
The memory and spd tabs will tell you what you have.

Lenovo 310 comes with amd and intel processors.
Both are capable of dual channel operation.
If you have amd, buy the exact product that Crucial suggested.
amd is tightly connected to ram.
Intel is much more tolerant of diverse ram.
If you do not have access to crucial ram or Kingston, buy a stick with the exact same specs that they recommend.

the memory says slot 1 and slot 2 both empty

Not possible.
Your link shows a single 4gb ram installed running in single channel mode.
If you select the spd tab, it should show slot#1 occupied and slot#2 empty.

If you have to buy a stick, buy 2133 speed and 15 timing.
Here is what crucial recommended:
https://www.newegg.com/crucial-16gb-260-pin-ddr4-so-dimm/p/N82E16820156081
To be safe, that is what I would buy.
I see other similar sticks that are lower priced. You take your chances there.
At least crucial will give you support if need be.
