I'm upgrading me PC and have narrowed down price range to Ryzen 5 1600. Will I need a cooling system for this CPU as I think the 1600x comes with cooling? If it is fine without cooling then I will just go with the 1600 which is £50 cheaper.
The R5 1600 comes with cooling the more expensive R5 1600X does not. So you will be fine and unless you plan on overclocking to 4Ghz the cooling solution that comes with the R5 1600 is very good and has been shown to allow for some overclocking (Most reviewers I saw were able reliably achieve 3.8ghz with it)
Do you think it is worth upgrading from the A10 7850k? Would it show a significant increase in gaming terms worth spending the money?
If the games are CPU limited then yes it should show a huge performance improvement. But this also depends on the GPU you are using and the resolution you play at. Offhand games I can think of that are CPU limited would be Skyrim, GTA V, Starcraft II, Civ 5/6, Ashes of the Singularity...

But otherwise it is very worth the upgrade the R5 1600 is a much much faster chip than the A10 7850K.
