does the generation if PCIE slot make a difference?

So does the generation of a pcie 16x slot make a difference on gaming performance for most graphic cards?
Or only matters when using something like a gtx titan?

If by generation you mean 2.0 vs 3.0 It has no affect unless you can fill the bandwidth of the slot. Such as a 780Ti, 690, or Titan.
