Mobo is a Gigabyte Gaming K5 Z270. Recently it has started beeping five times when starting up from being turned off. It delays the booting, but it eventually boots up. When I restart with the power on (warm boot), it POSTs fine and quickly boots up.

I think I have isolated an iffy RAM card (I run 32 GB in 4x8) — when I remove it, it doesn’t beep 5 times but starts normally. But my question is, why the difference between behavior in cold and warm boot? (Keep in mind that it seems to work fine after starting up either way, no instability, no BSODs.). Are the RAM tests more rigorous on cold boot, and it sees something is not quite right on one of the modules (even though it works)?

Windows 10’s memory diagnostic reported no errors.

We're going to need a little more information. Please include your system's specs like so:

Include the BIOS version you're on for the motherboard and the OS version. Can you state the age of the PSU as well?
