The drive is accessible right?
You should be able to access it and store some data over there.
You can start with just moving music, pictures, videos over there. You can do so with simple drag and drop (copy/paste).
As for your applications, you will need to uninstall them and reinstall them back on your E: drive. During the installation process, you will be asked where do you want to install the specific tool.
You have two separate, physical drives. So, you can expect that data after one of the drive is filled up, all the data would go to your secondary one.
