I use Edifier R1280T without the exterior grill. So the subwoofers are exposed to dust. It appears that the maid has cleaned them directly with the regular dusting clothes and that too with force. There's no holes or any sign of visible damage on the speakers. As I understand it, these things are extremely fragile, right? How would I know if perhaps the quality of the audio has been affected by rough cleaning? Would the sound get muddled suddenly?

Solution
I am talking about the membrane. Aren't the membrane/speaker supposed to be cleaned with a microfibre lint-free cloth? I just used a normal cloth.

Those are corrugated paper cone drivers that are 1.5X thicker than what was used in the 70's-80's, You are good using a normal cloth to brush away dust it will do no damage at all. Those will even resist a bit of water splashing since they are also treated before being compressed into shape.
Fragile? No, you can dust them without worry of damage but if you want to be sure then turn the volume to 50% and play this. If your voice coils are warped you will notice right off.
